The world No 321 recovered from three consecutive bogeys during his front nine to card a joint-low round-of-the-day 67 in Mississippi.
Armour, chasing a maiden PGA Tour title, birdied four of his final six holes to pull clear of nearest challenger Chesson Hadley.
Ireland’s Seamus Power is part of a five-way tie for third after a one-under 71, with Vaughn Taylor also part of the group on nine under.
Taking a two-shot lead into the weekend, Armour extended his advantage by birdieing the first and holing a 15-footer at the par-four next.
The American missed from eight feet to bogey the fifth and failed to get up-and-down from the bunker to save par at the next, before landing short of the par-three seventh green to post another blemish.

Armour picked up a shot at the eighth and converted from just off the 10th green for a birdie-two, with four straight gains from the 13th – including a 55-footer at the 16th – putting him in a commanding position.
Hadley birdied three of his first four holes to grab second, while Power and Taylor are joined six off the pace by Beau Hossler, Ben Silverman and Scott Strohmeyer.
World No 75 William McGirt, the highest ranked player in this week’s a field due to the WGC-HSBC Champions in China, sits tied-23rd after a two-under 70.
